چکیده

[14C]Cadaverine was applied to leaves of Lupinus polyphyllus, L. albus, L. angustifolius, L. perennis, L. mutabilis, L. pubescens, and L. hartwegii and it was preferentially incorporated into lupanine. In Lupinus arboreus sparteine was the main labelled alkaloid, in L. hispanicus it was lupinine. A pulse chase experiment with L. angustifolius and L. arboreus showed that the incorporation of cadaverine into lupanine and sparteine was transient with a maximum between 8 and 20 h. Only leaflets and chlorophyllous petioles showed active alkaloid biosynthesis, where as no incorporation of cadaverine into lupanine was observed in roots. Using in vitro organ cultures of Lupinus polyphyllus, L. succulentus, L. subcarnosus, Cytisus scoparius and Laburnum anagyroides the inactivity of roots was confirmed. Therefore, the green aerial parts are the major site of alkaloid biosynthesis in lupins and in other legumes.

Enzymatic Synthesis of Quinolizidine Alkaloids in Lupin Chloroplasts

The enzymatic sequence responsible for the biosynthesis of tetracyclic quinolizidine alkaloids could be localized in chloroplasts isolated from Lupinus polyphyllus leaves and L. albus seedlings by differential centrifugation. Upon feeding of cadaverine to isolated chloroplasts lupanine is produced as the main alkaloid. Chloroplasts treated with digitonine produce sparteine and 17oxosparteine in...

On the Role of A 1-Piperideine and Tripiperideine in the Biosynthesis of Quinolizidine Alkaloids

Carbon 14-labelled cadaverine, A 1-piperideine and a-tripiperideine were administered to leaf disks of Lupinuspolyphyllus in short term experiments of 12—20 h duration. Whereas cadaverine was readily incorporated into lupanine in good yields, a 7 to 60 times lower incorporation was observed for tripiperideine or A 1-piperideine. In L. arboreus the incorporation of cadaverine into sparteine was ...

Chemical Defense of Lupins. Mollusc-Repellent Properties of Quinolizidine Alkaloids

Polyphagous molluscs such as H elix pom atia and Arion rufus generally do not feed on plants containing alkaloids. O f 19 species tested 10 species were totally avoided, the other 9 species were less attacked than Lactuca sativa, which was readily taken. Plants containing quinolizidine alkaloids were studied in detail. Those species with the a-pyridone alkaloids cytisine and N-methylcytisine we...

Stereochemical aspects of the biosynthetic routes leading to the pyrrolizidine and the quinolizidine alkaloids

The regiochemistry and the stereochemistry of the biosynthetic sequences leading into retronecine from ornithine and putrescine, and into lupinine, sparteine and lupanine from lysine and cadaverine, is revealed by tracer experiments with intramolecularly doubly 13C,15N—labelled and chirally 2H—labelled substrates. The labelling patterns within the biosynthetic products were established by means...

Using elicitors for enhanced production of secondary metabolites in plant cell and organ suspension cultures

In biotechnology has been concentrated to review alternative routes for the production of natural compounds. Plant cell culture systems are viable alternatives for the production of secondary metabolites that are of commercial importance in food and pharmaceutical industries. However, relatively very few cultures synthesize these compounds over extended periods in amounts comparable to those fo...
